4-肼基苯甲酸及其酯可以作为医药和材料的中间体。在合成目标产物后,由于其含有羧基,可以通过对该基团进行进一步修饰来增加发现新药的机会。
制备 对氨基苯甲酸甲酯盐酸盐(3)的合成在1L烧瓶中加入500mL无水甲醇,并冷却于冰水浴中。缓慢滴加65mL氯化亚砜,控制内温不超过15℃。滴完后搅拌15分钟,然后一次性加入100g对氨基苯甲酸,加热回流3.5小时。冷却后析出白色固体,过滤并用冷甲醇洗涤,干燥得到中间体3,几乎为定量收率(136.1g)。
对肼基苯甲酸甲酯盐酸盐(5)的合成在1L三口烧瓶中加入300mL水,缓慢加入55mL浓盐酸,并加入50g中间体3。降温至-10℃以下,在此温度下滴加20g NaNO2溶液,控制反应液温度不超过0℃。当试纸检测反应完成后停止滴加,然后在低温下搅拌备用。
另一个1L三口烧瓶中加入280mL水,并搅拌下加入87g Na2SO3使其溶解。用浓盐酸调节Na2SO3溶液pH值至6.5~6.8。将上述重氮化溶液缓慢地呈细线状滴加到Na2SO3溶液中,控制反应液温度不超过20℃。加完后,在室温下搅拌0.5小时,再加入20ml浓盐酸升温到90℃回流3小时,自然降至室温,在10℃下搅拌0.5小时,抽滤并用20mL 10%(质量分数)盐酸洗涤,冰水洗涤后减压真空干燥得到黄色固体48.6g,收率90%,熔点为233~235℃。
4-肼基苯甲酸(1)的合成在500mL烧瓶中加入30g中间体5和200mL1:1(V/V)乙醇-水,搅拌下缓慢滴加50%NaOH溶液40ml,在氮气保护下升温回流至无原料为止,约需3小时。回收乙醇至尽,冷却到室温后用浓盐酸调节反应液pH值为4~4.1,并在10℃下搅拌1小时。过滤并用少量冰水洗涤,真空干燥得到黄色固体25.5g,收率91%。
