Iridium‐Catalyzed Direct CH Amidation with Weakly Coordinating Carbonyl Directing Groups under Mild Conditions

An iridium‐catalyzed direct CH amidation of weakly coordinating substrates, in particular of those bearing ester and ketone groups, under very mildconditions has been developed. The observed high reaction efficiency was achieved by the combined use of acetic acid and lithium carbonate as additives.
The present invention relates to novel tyrosine alkoxyguanidine compounds that are inhibitors of alpha V (&agr;v) integrins, for example &agr;v&bgr;3 and &agr;v&bgr;5 integrins, their pharmaceutically acceptable salts, and pharmaceutical compositions thereof. The compounds may be used in the treatment of pathological conditions mediated by &agr;v&bgr;3 and &agr;v&bgr;5 integrins, including conditions such as tumor growth, metastasis, restenosis, osteoporosis, inflammation, macular degeneration, diabetic retinopathy, and rheumatoid arthritis. The compounds have the general formula:

Novel and efficient synthesis of S-aryl/alkyl phosphinothioates from P(O)–H and aryl/alkyl sulfonylchlorides under metal- and base-free conditions is described. This reaction provides an alternative strategy for the construction of P–S–C bonds in moderate to excellent yields. Moreover, this method can be readily applied to gram-scale preparation.

A cheap, easy‐to‐execute and sustainable synthetic methodology for the synthesis of indoles and benzofurans was developed utilizing Pd/C as the promoter. A great variety of substituted ortho‐allyl anilines or phenols were utilized, leading to products in moderate yields. Recycling of the catalyst up to five times has been achieved.
