Skin contact: Immediately wash skin with copious amounts of water for at least 15 minutes while removing

概述

制备 步骤1:2-氨基-4-溴苯甲醇的合成

将60g NaBH4加入300mL四氢呋喃(THF)中,机械搅拌。在冰水浴条件下,向溶液中分批加入100g 2-氨基-4-溴苯甲酸,并缓慢滴加27mL三氟化硼乙醚溶液,保持反应温度低于20℃。反应完毕后将溶液升至室温并过夜,直至原料完全反应。随后,向反应液中依次加入300ml甲醇和5N NaOH(200ml),搅拌约半小时。通过旋转蒸发除去有机溶剂,并冷却至室温,析出固体进行过滤,得到2-氨基-4-溴苯甲醇粗品约60g[质谱(MS)M+1 203]。收率98.7%。

步骤2:2-氨基-4-溴苯甲醛的合成

将步骤1获得的产物60g和300g二氧化锰加入1L二氯甲烷溶剂中,机械搅拌反应过夜。然后通过硅藻土过滤,并经石油醚与乙酸乙酯(5:1)混合液进行柱层析得到2-氨基-4-溴苯甲醛约40g[核磁共振氢谱(400MHz,CDCl3):δ6.72-6.75(d,1H),7.23(bs,2H),7.38-7.42(dd,1H),7.72-7.73(d,1H),9.77(s,1H)]。收率85.2%。
