    In some embodiments, the present disclosure relates to mutated tRNAs in which the first letter of the anticodon has been substituted to lysidine or agmatidine, and translation systems containing the mutated tRNAs. In a specific embodiment, the present disclosure provides mutated tRNAs capable of selectively translating codon NNA. In another embodiment, the present disclosure provides translation systems capable of translating two or three types of amino acids from a single codon box. In still another embodiment, the present disclosure provides novel methods for synthesizing lysidine diphosphate, agmatidine diphosphate, and derivatives thereof.

    An objective of the present invention is to provide novel means that allow reducing the rate of mistranslation into unintended amino acids due to misreading of codons. In a number of embodiments, the present disclosure provides compositions for translation which contain a tRNA having a specific combination of bases at positions 32, 33, 37, and 38 (tRNA numbering rule) in the tRNA. According to a specific embodiment of the present disclosure, the rate of introduction of unintended amino acids due to codon misreading can be reduced.

    Non-naturally occurring tRNASec and methods of using them for recombinant expression of proteins engineered to include one or more selenocysteine residues are disclosed. The non-naturally occurring tRNASec can be used for recombinant manufacture of selenocysteine containing polypeptides encoded by mRNA without the requirement of an SECIS element. In some embodiments, selenocysteine containing polypeptides are manufactured by co-expressing a non-naturally occurring tRNASec a recombinant expression system, such as E. coli, with SerRS, EF-Tu, SelA, or PSTK and SepSecS, and an mRNA with at least one codon that recognizes the anticodon of the non-naturally occurring tRNASec.
