Psilostachyin, acetylated pseudoguaianolides and their analogues: Preparation and evaluation of their anti-inflammatory potential

Naturally occurring acetylated pseudoguaianolides and psilostachyin including their analogues were synthesized. The structure of semi-synthetic psilostachyin was also confirmed by X-ray crystallography. The anti-inflammatory potential of all the derivatives has been evaluated through in vitro expression of TNF-alpha, IL-1 beta and IL-6 in murine neutrophils. (C) 2011 Elsevier Ltd. Parthenin and some of its derivatives have been shown to posses significant anti-malarial activity against a multi-drug resistant strain of Plasmodium falciparum. Structure-activity relationships in this series of compounds are discussed and possible mechanisms of action considered. Molecular matching of the oxygen atoms of parthenin and the new anti-malarial drug qinghaosu shows both molecules to be amphipathic with common areas for their oxygen atoms and hydrocarbon frameworks.
