  本发明属于有机化合物工艺应用技术领域，公开了一种构建立体和大位阻二硫醚化合物(III)的合成方法，在反应溶剂中，以N‑二硫邻苯二甲酰亚胺(I)和具有alpha氢的酯(II)为反应原料，以路易斯碱或者金鸡纳碱为催化剂，在特定温度条件下，高收率得到多种具有季碳中心的立体和大位阻二硫醚化合物(III)。本发明合成方法原料易得价廉，反应操作简单，反应条件温和，产率较高，官能团耐受性良好。本发明实现了一些药物分子，生物小分子，荧光分子的二硫连接和修饰，得到了一些手性二硫醚化合物，为药物化学特别是ADC药物的连接提供了一种快速稳定的连接方式，有望在药物化学领域得到广泛应用。
