Symmetrical α-bromoacryloylamido diaryldienone derivatives as a novel series of antiproliferative agents. Design, synthesis and biological evaluation

In a continuing study of hybrid compounds containing the alpha-bromoacryloyl moiety as potential anticancer drugs, we synthesized a novel series of hybrids 4a-h, in which this moiety was linked to a 1,5-diaryl-1,4-pentadien-3-one system. Many of the conjugates prepared (4b, 4c, 4e and 4g) demonstrated pronounced, submicromolar antiproliferative activity against four cancer cell lines. Moreover, compound 4b induced apoptosis through the mitochondrial pathway and activated caspase-3 in a concentration-dependent manner. (C) 2010 Elsevier Ltd. All rights reserved.

A selective mono-1,4-hydrocyanation of 10 diarenyl ketones usingpotassiumhexacyanoferrate(II) as an eco-friendlycyanidesource, potassium hydroxide as a catalyst, and benzoyl chloride as a promoter has been achieved. This protocol has the advantages of a non-toxic cyanidesource, a heavy metal-free catalyst, selectivity, very good yields and a simple work-up procedure.

A catalytic aldol condensation in the presence of lithium perchlorate and tertiary amines is described giving pure products in high yields. The aldol condensation can be performed even in the presence of hydrated lithium perchlorate.

yrazole (1) with substituted diarylidenketones (2) constitutes a convenient synthetic route to the hitherto unknown 1-benzyl-6-(4-chlorophenyl)-2-(4-R-phenyl)-4-(4-R-styryl)-2,3-dihydropyrazolo-[3,4-b][1,4]diazepines (3). Structures of all products were consistent with their IR, 1H-NMR, 13C-NMR and MS spectral data. X-ray crystallography data confirm the assigned structures.
