    The 8'-benzophenone, 8'-dansylhydrazone, 3'-S-(2-ethyldansylamide), and 3'-S-acetamidofluorescein derivatives of the plant hormone abscisic acid (ABA) have been synthesized for use in photoaffinity labeling (the benzophenone derivative) or fluorescence probe experiments and have been spectroscopically characterized. One of the three fluorescent compounds, the 3'-tethered fluorescein derivative, exhibits spectroscopic and photophysical properties which indicate that it could be an excellent fluorescent probe of ABA interactions in vivo. The 3'-tethered fluorescein and ABA moieties do not interact strongly, so that the fluorescence properties of the fluorescein-labelled hormone are very similar to those of fluorescein itself. Measurements of the absorption, emission, and fluorescence excitation spectra, fluorescence quantum yields, and fluorescence decay parameters of this derivative as a function of pH indicate that the photophysics is dominated by ground and excited state prototropic equilibria involving only the fluorescein moiety. The fluorescein dianion is the only significant absorber and emitter at pH > 6.7, whereas only the cation absorbs and emits at pH < 0. In the intervening pH range, strong emission from the monoanion and weak emission from two neutral species, tentatively assigned to the zwitterion and the lactone of the fluorescein moiety, is observed.Key words: abscisic acid, fluorescein, synthesis, photophysics.

  • Activity and utility of abscisic acid having a 3′ thioether linker arm

    Abscisic acid (ABA) was functionalized at the 3'-position by reaction of its alpha-2',3'-epoxide with 1,4-butanedithiol to afford the corresponding derivative having a six-atom linker arm containing a terminal sulphhydryl group. This derivative was coupled with epoxy activated Sepharose 6B affording a gel containing ca 1.4 mg of (+)-ABA ml(-1). This gel was shown to bind specifically an anti-ABA antibody from hybridoma culture broth, and its binding capacity was determined. The (+)-ABA analogue possessing the thiol linker arm was also derivatized by reaction with a fluorescent derivatizing agent and these compounds were shown to inhibit cress seed germination.     (+)-3'-Chloro, -bromo and -iodoabscisic acids were synthesized, and their biological activities and metabolism were examined, as well as that of (+)-3'-fluoroabscisic acid. This was in order to estimate the effects of altered electron density at the C-2' position of abscisic acid on both biological activity and metabolic stability. The biological activities of (+)-3'-chloro acid bromoabscisic acids were similar to, or higher than, those of(+)-abscisic acid in four bioassays, while (+)-3'-iodoabscisic acid was more potent in two bioassays and less potent than (+)-abscisic acid in the other two bioassays. The biological activities of 3'-haloabscisic acids seem to be dependent on the 3'-halogens, but not on the electron density at C-2'. The metabolism of(+)3'-haloabscisic acids to ca. 50% of their original levels in the media of rice cell suspension culture was similar to that of(+)abscisic acid. Metabolites from (+)-3'-chloro, -bromo and -iodoabscisic acids were not found in the culture media and cells, while (+)-3'-fluoroabscisic acid gave (+)-3'-fluoro-8'-hydroxyabscisic acid as a stable metabolite. (+)-3'-fluoro-8'- hydroxyabscisic acid showed biological activities lower than (+)-3'-fluoroabscisic acid, but higher than (-)-3'alpha-Fluorophascsic acid. These findings suggested that the biological activity of (+)-8'-hydroxyabscisic acid is intermediate between those of (+)abscisic acid and (-)-phaseic acid.     (S)-(+)-3′-Fluoroabscisic acid (5a) was synthesized as an analogue of abscisic acid that can resist the metabolic cyclization of 8′-hydroxyabscisic acid to phaseic acid due to increased electron density at C-2′.
