Certain alkyl amino-glucopyranoside are disclosed, including alkyl N-carbamyl-N'-(2-chloroethyl)-N'-nitroso-6-amino-6-deoxy-D-glucopyranoside , alkyl N-carbamyl-N'-(2-chloroethyl)-N'-nitroso-2-amino-2-deoxy-D-glucopyranoside , and alkyl di-N,N'-[N-(2-chloroethyl)-N-nitroso-carbamyl]-2,6-di-amino-2,6-di-deoxy-D -glucopyranoside. The alkyl amino-glucopryanoside exhibits marked antitumor activity while not having diabetogenic activity and bone marrow toxicity. The alkyl amino-glucopryanoside is further water soluble and free from antibacterial activity.

3-cyclohexanedione in MeOH in the presence of Et3N afforded 2-deoxy-2-[(4,4-dimethyl-2,6-dioxocyclohexylidenemethyl)amino]- d -glucose (6) in yields > 75%. A new class of nitrosoureas. VI. Synthesis and antitumor activity of 3-substituted-1-(2-chloroethyl)-3-(methyl-.ALPHA.-D-glucopyranosid-2-yl)-1-nitrosoureas.
A series of five 3-substituted-1-(2-chloroethyl)-3-(methyl-α-D-glucopyranosid-2-yl)-1-nitrosoureas (5a-e) was prepared and tested for antitumor activities. The compounds were obtained by the reaction of N-substituted methyl-α-D-glucosaminides (3a-e) with isocyanate followed by nitrosation with dinitrogen tetroxide. All the nitrosoureas obtained were remarkably active against leukemia L1210 and Ehrlich ascites carcinoma and showed greater therapeutic ratios than the positive controls ; 1-(2-chloroethyl)-3-(β-D-glucopyranosyl)-1-nitrosourea (GANU) and 1-(2-chloroethyl)-3-(D-glucopyranos-2-yl)-1-nitrosourea (DCNU).
