制备方法与用途

羧甲基壳聚糖是一种水溶性壳聚糖衍生物,为一种两性聚电解质,其是在壳聚糖高分子链上引入亲水基团-CH2COOH而成,壳聚糖经羧甲基化改性后,提高了水溶性,尤其是在中性和碱性溶液中的溶解性显著增强,使其具有成膜、增稠、保湿、螯合等的特性,所以其在农业、医药、保健品、化妆品、保鲜、环保等方面有广泛应用。

羧甲基壳聚糖(CMC)为壳聚糖的羧甲基化衍生物,其极性较强、易溶于水,已广泛用于制药、生物医用材料开发等领域。由于药物中羧甲基壳聚糖含量会直接影响其抗菌、抗感染的临床作用效果,因此对羧甲基壳聚糖进行准确定量显得尤为重要。

在反应瓶中加入氢氧化钠的异丙醇溶液中,氢氧化钠的浓度为40%。

将脱乙酰度为15%~45%的壳聚糖加入反应瓶中,加入后壳聚糖与氢氧化钠的质量比为1:4.8。

将预先溶解好的氯乙酸的异丙酮溶液在搅拌状态下,缓慢滴加到反应瓶中,加毕继续搅拌反应3h。壳聚糖与氯乙酸的质量比为5:1。

反应完毕后向反应后物料中加入浓盐酸,调节物料pH值为6~7,抽滤,用无水乙醇洗涤滤饼3次,洗涤后的滤饼在50℃以下干燥后得到羧甲基壳聚糖粗品。

纯化羧甲基壳聚糖的方法,包括如下步骤:

1)将等电点时pH为2.7,数均分子量为40W,羧甲基取代度为1.3的40Kg羧甲基壳聚糖用2吨水溶解后,将羧甲基壳聚糖水溶液的pH调至2.7,使羧甲基壳聚糖形成白色絮状沉淀,离心脱水得到浓缩羧甲基壳聚糖;

2)用pH为2.7的过滤水反复洗涤1)中获得的浓缩羧甲基壳聚糖,去除浓缩羧甲基壳聚糖中所含盐分,获得去盐羧甲基壳聚糖;

3)用浓度为95%的乙醇反复洗涤所述的去盐羧甲基壳聚糖至上层清液的酒精度数为95°,去除上层清液离心干燥获得纯度为99.3%的羧甲基壳聚糖。

完成以上纯化过程,总共用到了430Kg浓度为95%的乙醇,与现有技术中纯化40Kg同样的羧甲基壳聚糖需要2吨95%的乙醇相比,大大节省了乙醇的用量,降低了生产成本,减少了废酒精的排放,避免了对环境的破坏。
