4-氯苯乙醇是我国规定允许使用的食用香料,可用于调配玫瑰香型花精油和各种花香型香精,如茉莉香型、丁香香型、橙花香型等。它广泛用于调配皂用和化妆品香精。
合成方法将装有搅拌棒的100 mL烘箱干燥圆底烧瓶置于氩气正压下,并在高真空下进行三次排空/回填循环。加入碘化钐(II)(6当量,THF溶液,浓度为0.080 M),然后依次加入乙基三甲基胺(Et3N)和水(均为36当量)。将癸酰胺的THF储备溶液(1.0当量,1.0 mmol,体积5.0 mL)添加到预制的碘化钐(II)/胺/H2O络合物中,并将反应混合物搅拌18小时。通过向反应混合物中通入空气气泡来氧化过量的SmI2。用CH2Cl2(100 mL)和NaOH(50 mL,1 N)稀释反应混合物。用 (2 x 100 mL)萃取水层,并合并有机层。经Na2SO4干燥、过滤并浓缩后,粗反应混合物的分析显示转化率大于98%,选择性大于98:2。通过色谱纯化得到癸-1-醇。特征数据包含在后续章节中。
2-(4-氯苯基)乙酰胺(表2,条目12),按照一般程序,在用 /NaOH(1.0 N)处理并以85%的产率层析后得到标题化合物。最终产物为4-氯苯乙醇。
