    Divergent Synthesis of All Possible Optically Active Regioisomers ofMyo‐Inositol Mono‐ and Bisphosphates
    摘要:
    All possible optically active regioisomers of myo- inositol mono- and bisphosphates were synthesized using inositol derivatives suitably protected with various protecting groups (IR(n)s) as key intermediates. A series of procedures including Novozym 435 catalyzed enzymatic resolution of (3aR, 4S, 7S, 7aR)-rel-3a, 4,7,7a-tetrahydro- 2,2-dimethyl- 1,3- benzodioxole-4,7- diol diacetate, several protection and deprotection reactions, and acyl migration afforded two enantiomeric pairs of IR5 and six enantiomeric pairs of IR4. Phosphorylation of these key intermediates by the phosphitylation and oxidation procedure gave the target products after removal of the protecting groups.[GRAPHICS]

    The invention provides diazo compounds having the formula XYCNN and their parent hydrazones, where X is an ortho-nitro aromatic group such as o-nitrophenyl and Y is a hydrocarbon group such as methyl. The hydrazones may be prepared by reacting the corresponding ketone with hydrazine and the diazo compounds by hydrazone oxidation. The diazo compound may be reacted with an organic compound having a reactive hydrogen atom such as phosphate, thiophosphate, phosphonate, carboxyl and phenol, to form a caged compound from which the original organic compound can be released by photolysis, eg in situ in a biological system. Preferred organic compounds are ATP and other nucleotides.

    An insulin activity messenger material and its precursor material are disclosed which have been isolated from hapatic plasma membrane cells that have been incubated with an enzyme known as a phosphatidylinositol-specific phospholipase C. The messenger material comprises a carbohydrate-based compound that exhibits the ability to modulate the activity of the insulin-sensitive enzymes pyruvate dehydrogenase, adenylate cyclase, acetyl CoA carboxylase, and low Km CAMP phosphodiesterase, and thereby effectuate the activity of insulin on the cellular level. The precursor material comprises an inositol containing glycolipid capable of phosphodiesteratic cleavage by a phosphatidylinositol-specific phospholipase C. The enzyme phosphatidylinositol-glycan specific phospholipase C has also been purified and found to participate in many of the activities identified with respect to the messenger and its precursor. Numerous diagnostic and therapeutic utilities are proposed, and testing procedures, materials in kit form and pharmaceutical compositions are likewise set forth.
