抗代谢类抗肿瘤药

抗代谢药是一类与正常代谢物或辅酶结构相似的化合物,主要通过与合成正常代谢物所必需的酶结合,从而干扰核酸的合成,抑制肿瘤细胞的生长和增殖。某些抗代谢物还具有反馈抑制作用,能够阻断代谢物前体的合成。常见的抗代谢药包括抗叶酸、抗嘌呤及抗嘧啶等,它们主要作用于细胞周期的S期,属于典型的细胞周期特异性药物。

由于肿瘤组织与正常组织在代谢过程上的差异不大,因此这类药物的选择性较差,对诸如骨髓、胃肠道黏膜、生殖系统和毛发等增殖较快的正常组织也具有一定的毒性。尽管多数抗代谢药主要抑制细胞DNA合成,但由于它们的作用点不同,各药物之间通常不会产生交叉耐药性。
