  摘要：

  1.用人类肝脏在体外研究了澳大利亚土著药用植物多多纳多萝（Dodonaea polyandra）的成分消炎性二萜类多聚酸A（PAA）的代谢及其去酯化的醇代谢产物水解多聚酸A（PAAH）。微粒体（HLM）和重组UDP-葡萄糖醛酸转移酶（UGT）和细胞色素P450（CYP）酶。 2.与HLM孵育后，发生PAA水解以生成PAAH。在UGT和CYP辅因子存在下，PAAH与HLM进一步孵育会导致大量耗竭，其中UGT介导的耗竭是主要途径。 3.利用选择性酶抑制剂和重组人UGT和CYP酶的反应表型揭示了UGT2B7和UGT1A1，以及CYP2C9和CYP3A4是参与PAAH代谢的主要酶。 4.通过UPLC / MS / MS分析，用UDP-葡糖醛酸补充的HLM和重组酶对PAAH的温育，鉴定出三种葡糖醛酸代谢物。代谢物的进一步特征是β-葡萄糖醛酸苷酶和温和的碱性水解。已证明PAAH的酰基葡糖醛酸苷是主要的代谢产物。
