First catalytic enantioselective synthesis of P-stereogenic phosphoramides via kinetic resolution promoted by a chiral bicyclic imidazole nucleophilic catalyst

A novel method toward the synthesis of P-stereogenic organophosphorus compounds via kinetic resolution by a catalytic asymmetric phosphorylation through P-N bond formation has been developed. Using a chiral bicyclic imidazole nucleophilic catalyst, chiral phosphoramide compounds were prepared in high conversions and with moderate enantioselectivity. This is also the first example of the catalytic asymmetric synthesis of chiral phosphoramide compounds. (C) 2012 Elsevier Ltd. All rights reserved.

The present invention features compositions and methods for the treatment of HCV infection. In one embodiment, the compositions of the invention comprise (1) Compound 1 or a pharmaceutically acceptable salt thereof, and (2) Compound 2a or a pharmaceutically acceptable salt thereof. In another embodiment, the compositions of the invention comprise (1) Compound 1 or a pharmaceutically acceptable salt thereof, and (2) a prodrug of Compound 2a (e.g., one of Compounds 2b-2k or Examples 3-1 to 3-10) or a pharmaceutically acceptable salt of said prodrug.

A new type of chiral bicycle imidazole nucleophilic catalyst was rationally designed, facilely synthesized, and successfully applied in an asymmetric Steglich rearrangement with good to excellent yield and enantioselectivity at ambient temperature. Moreover, it can be easily recycled with almost no reduction of catalytic efficiency. This is the first example for the successful chiral imidazole nucleophilic
