Total synthesis of lycoperdic acid and its C4-epimer

Efficient stereodivergent syntheses of (+)-lycoperdic acid (LPA) and 4-epi-LPA have been achieved based on asymmetric hydrogenation (H2, Rh/(R,S)-MeBoPhoz) of racemic enamide as a key step.
Mushroom-derived glutamate natural product lycoperdic acid and its three stereoisomers were synthesized in 1.2–7.1% yield for total 11 steps from 2,2-dimethyl-1,3-dioxan-5-one. This paper describes the second half of these synthetic works via a suitably protected amino acid intermediate with stereochemically defined complete carbon chain each. The neuronal activities are also reported.
