A 2+3 condensation of a wide assortment of Schiff bases, prepared from aromatic aldehydes and primary amines, with methyl 2-(1-ethoxyethylideneamino)acetate allows convenient access to an extensive family of substituted 4-arylideneimidazolin-5-one analogues of the green fluorescent protein (GFP) chromophore. 4-arylideneimidazolin-5-one - azomethine ylide - Schiff bases - heterocycle - 2+3 cycloaddition
chromophore-based fluorescentprobe (BDI-ER) was designed and prepared for selectively labelingendoplasmicreticulum. The core chromophore of BDI-ER is HBDI which derived from the core structure of GFP. And two hydrophobic octadecane chains was anchored on the two tails of the BDI-ER, which allows the amphipathic nature of the probe. BDI-ER exhibited high selectivity for labelling ER in various cell lines.

This work discovered that fluorescence protein chromophore analogs can undergo triplet excitation, enabling photo-crosslinking and photo-sensitizing chemistries. Exploration of these syntheticfluorescent protein chromophores resulted in photo-crosslinkers for biomolecules, photo-catalysts for organic reactions, and photosensitizers for chromophore-assisted light inactivation and photodynamic therapies
