    Identification of an acridine photoaffinity probe for trypanocidal action
    摘要:
    Twenty-four acridine derivatives were screened for trypanocidal activity in Trypanosoma brucei in order to determine which structural features of the acridine molecule confer maximal antiparasitic activity. The synthesis of several new azidoacridine derivatives are also reported as well as an assessment of their value as possible photoaffinity probes for the study of acridine trypanocidal action. The most effective and selective acridine trypanocides, with and without irradiation, were the 3-amino-10-methylacridinium salt derivatives. With brief irradiation, one azidoacridine, 3-amino-6-azido-10-methylacridinium chloride, showed considerable trypanocidal activity at very limiting drug concentrations (10(-7)M) and warrants consideration as a possible photoaffinity probe.

    The invention relates to novel fluorescent dyes with multiple negatively charged groups in their ionized form which are 9-aminoacridines or 1-aminopyreneshaving of one of the following general formulae A-E: or salts or protonated forms thereof, wherein the ionizable groups are typically selected from the following: OH, SH, COOH, SO3H, OSO3H,SO2NHCN, P(O)(OH)2, P(O)(OH)2. The invention further relates to the use of these dyes as fluorescent tags, in particular for reducing sugars and glycans, and to carbohydrate-dye conjugates comprising these dyes as well as to methods for preparing the same.

    A novel set of dimers derived from 9-amino acridine was prepared and characterized by H-1 and C-13 NMR. These derivatives are bridged at several different positions of the heterocyclic moieties, by the way of alpha,omega-diamido or alpha,omega-diamino side-chains. Additionally the preparation of some bi-bridged compounds was achieved.
