    Structure–activity relationship study of acridine analogs as haspin and DYRK2 kinase inhibitors
    摘要:
    Haspin is a serine/threonine kinase required for completion of normal mitosis that is highly expressed during cell proliferation, including in a number of neoplasms. Consequently, it has emerged as a potential therapeutic target in oncology. A high throughput screen of approximately 140,000 compounds identified an acridine analog as a potent haspin kinase inhibitor. Profiling against a panel of 270 kinases revealed that the compound also exhibited potent inhibitory activity for DYRK2, another serine/threonine kinase. An optimization study of the acridine series revealed that the structure-activity relationship (SAR) of the acridine series for haspin and DYRK2 inhibition had many similarities. However, several structural differences were noted that allowed generation of a potent haspin kinase inhibitor (33, IC(50) <60 nM) with 180fold selectivity over DYRK2. In addition, a moderately potent DYRK2 inhibitor (41, IC(50) <400 nM) with a 5.4-fold selectivity over haspin was also identified. (C) 2010 Elsevier Ltd. All rights reserved.

    Acridinium-Based Photocatalysts: A Sustainable Option in Photoredox Catalysis

    The emergence of visible light photoredox catalysis has enabled the productive use of lower energy radiation, leading to highly selective reaction platforms. Polypyridyl complexes of iridium and ruthenium have served as popular photocatalysts in recent years due to their long excited state lifetimes and useful redox windows, leading to the development of diverse photoredox-catalyzed transformations. The low abundances of Ir and Ru in the earth's crust and, hence, cost make these catalysts nonsustainable and have limited their application in industrial-scale manufacturing. Herein, we report a series of novel acridinium salts as alternatives to iridium photoredox catalysts and show their comparability to the ubiquitous [Ir(dF-CF3-ppy)(2)(dtbpy)](PF6).

    Novel crown ethers incorporating one to three 9-phenyl­acridinium units were synthesized in moderate yields. The acridinium units within the macrocycles were converted into the photoactive N-methyl-9-methoxy-9-phenyl-9,10-dihydroacridine (N-methyl-9-methoxy-9-phenylacridane) units.

    and environmental-friendly method for the construction of acridone derivatives with isatins and arynes was described. The isatins underwent a ring expansion process oxidized by O2 in the air and a broad scope of acridones in moderate to good yields were obtained. Baeyer-Villiger-type oxidation and intermolecular-nucleophilic addition were included, and no extra oxidant except O2 was needed.
