    Discovery and Optimization of Novel, Selective Histone Methyltransferase SET7 Inhibitors by Pharmacophore- and Docking-Based Virtual Screening

    Histone methyltransferases are involved in various biological functions, and these methylation regulating enzymes' abnormal expression or activity has been noted in several human cancers. Within this context, SET domain-containing (lysine methyltransferase) 7 (SET7, also called KMT7, SETD7, SET9) is of increasing significance due to its diverse roles in biological functions and diseases, such as diabetes, cancers, alopecia areata, atherosclerotic vascular disease, HIV, and HCV. In this study, DC-S100, which was discovered by pharmacophore- and docking-based virtual screening, was identified as the hit compound of SET7 inhibitor. Structure-activity relationship (SAR) analysis was performed on analogs of DC-S100 and according to the putative binding mode of DC-S100, structure modifications were made to improve its activity. Of note, compounds DC-S238 and DC-S239, with IC50 values of 4.88 and 4.59 mu M, respectively, displayed selectivity for DNMT1, DOT1L, EZH2, NSD1, SETD8, and G9a. Taken together, DC-S238 and DC-S239 can serve as leads for further investigation as SET7 inhibitors and the chemical toolkits for functional biology studies of SET7.

    An insoluble azo pigment is produced by ejecting a diazo solution and a grounding solution continuously into an ejector such that within the ejector, the diazo solution and grounding solution are mixed with each other and are subjected to a coupling reaction. A production system for the insoluble azo pigment includes the following units (1) to (4): (1) a first feed tank ( 2 ) for a first fluid selected from the diazo solution or grounding solution, a first flow path (a) for feeding the first fluid to nozzles of an ejector ( 1 ), and a pump (d) for feeding the first fluid through the first flow path; (2) the ejector ( 1 ) provided with the nozzles, a suction chamber, a diffuser, and a temperature control device (g); (3) a second feed tank ( 3 ) for a second fluid, a second flow path (b) for guiding the second fluid into the suction chamber, and a flow control device (e) for controlling a flow rate of the second fluid through the second flowpath (b); and (4) a heat treatment tank ( 5 ) for subjecting to heat treatment a suspension of the insoluble azo pigment formed by a reaction between the first fluid and the second fluid, and a third flow path (a′) for guiding the suspension from the diffuser to the heat treatment tank.

    Method of producing a disazo pigment, in which a coupling reaction is carried out while feeding, into an acidic aqueous solution, a tetrazo aqueous solution containing a tetrazo component of benzidines and a coupler aqueous solution containing a coupling component, wherein the coupling component is a mixture comprising a main component represented by the formula (1): CH3COCH2CONH-X, wherein X represents a phenyl group optionally having one or the same or different plural substituents selected from the group consisting of a methyl group, methoxy group and chlorine atom; a first additional component represented by the formula (2): CH3COCH2CONH-Y, wherein Y represents a phenyl group having a substituent selected from the group consisting of -CONR2 group, -SO2NR2 group and -NHCOR group, and the phenyl group may further have a substituent selected from the group consisting of a methyl group, methoxy group, chlorine atom, -CONR2 group, -SO2NR2 group and -NHCOR group, and wherein Rs may be the same or different mutually, and represent a hydrogen atom, C1 to C4 alkyl group (the alkyl groups may be mutually connected to form a ring) or C1 to C4 alkylene NR'2, further wherein R's represent a hydrogen atom or C1 to C4 alkyl groups which may be mutually different (the alkyl groups may be mutually connected to form a ring); and, a second additional component represented by the formula (3): CH3COCH2CONH-Z, wherein Z represents a phenyl group substituted by any one of a carboxyl group or alkali metal salts thereof, hydroxyl group or sulfonic acid group or alkali metal salts thereof, and said phenyl group may further has a substituent selected from the group consisting of a methyl group, methoxy group, chlorine atom, carboxyl group or alkali metal salts thereof, hydroxyl group or sulfonic acid group or alkali metal salts thereof. According to the method, disazo pigments can be obtained which manifest excellent transparency and flowability of the inks produced therefrom, and excellent flushing property and flushing drain coloration resistance in the pigment production process, or excellent dispersibility when ink is formed under dry condition.
