噻吩类化合物是仅含有一个硫原子的五元杂环化合物,基本骨架为噻吩。这种化合物主要存在于煤焦油和页岩油中。
相较于苯,噻吩更为活泼。它具有光敏性,在光照条件下会发生碳架重排;可以与金属反应;在阮内镍的作用下可脱除硫原子;还能够进行取代反应(多发生在2位);可以氧化开环,并生成不同产物;可以在2、5位发生硝化,形成硝基噻吩;卤化后可产生2位单卤化物、2,5-位二卤化物、2,3,5-三卤化物及四卤化物;与苯缩合能生成苯并噻吩。
由于硫的非共用电子对与两个碳原子结合,噻吩环形成了一个6π电子系统,并表现出芳香族性质。因此它可以进行硝化、磺化和卤化等亲电取代反应,取代位置通常为2-位和5-位。这类反应比苯更容易发生。例如,在温和的缩合剂如正磷酸存在下,噻吩即可发生Friedel-Crafts反应,在α-位引入酰基。
在硝酸作用下,与碘反应可以高产率制备2-碘噻吩;Mannich反应也可用于合成2-噻吩甲胺。
噻吩广泛应用于生产染料、香料、耐急冷急热塑料、高活性溶剂、刺激素、杀虫剂、增亮剂、化妆品以及生物活化物质和维生素、麻醉剂及抗菌素等药物。尽管可以通过化学方法制备,但由于成本较高,实际应用中噻吩主要从粗苯的预精馏轻苯中提取。
在轻苯酸洗精制过程中,噻吩会与硫酸反应生成噻吩磺酸,并溶解于废硫酸中;通过澄清、除去焦油状物质及水解蒸馏处理后,可得到含噻吩和苯族烃的粗馏油。进一步碱中和至中性或微碱性后,使用理论板数为30~40的精馏塔进行精馏,可以分离出高纯度的噻吩产品。而剩余残液则可用于精馏出间-二甲苯约95%含量的产品。
