Chemical syntheses of two disaccharides, benzyl beta-D-glucopyranosyl-(1 --> 2)-alpha-L-arabinopyranoside (1) and benzyl beta-D-glucopyranosyl-(1 --> 4)-alpha-L-arabinopyranoside (2), and a trisaccharide, benzyl beta-D-glucopyranosyl-(1 --> 2)-3-O-acetyl-4-O-(beta-D-glucopyranosyl)-alpha-L-arabinopyranoside (3), related to oat root triterpenoid saponin Avenacin A-1 are reported. Sulfuric acid immobilized on silicagel has been used as an efficient and safe alternative promoter for acetalation and subsequent acetylation of sugar glycosides using stoichiometric reagents without work-up. The synthesis of different types of per-O-acetylated acetals/ketals has been achieved from various types of O- and S-glycosides in excellent yields.

The first total synthesis of pulsatilla saponin D, a potent anti-lung cancer natural product was accomplished in eight steps from l-arabinopyranoside in high overall yield via a highly stereospecific glycosylation featuring an oxocarbenium mechanism between the trisaccharide derivative and aglycone as the key step.
La(OTf)3: An Efficient Promoter for Thioglycoside Activation in Conjunction with N-Iodosuccinimide

Use of La(OTf)3 as a Lewis acid promoter for N-iodosuccinimide-mediated activation of thioglycosides is reported. The glycosylation reactions proceeded smoothly with good to excellent yields and stereoselectivity.
