Presently provided are inhibitors of cellularly expressed TDO2 and pharmaceutical compositions thereof, useful for modulating an activity of tryptophan 2, 3 dioxygenase; treating immunosuppression; treating a medical conditions that benefit from the inhibition of tryptophan degradation; enhancing the effectiveness of an anti-cancer treatment comprising administering an anti-cancer agent; and treating tumor-specific immunosuppression associated with cancer.
The preparations of cyclopropanone (1), methylcyclopropanone (2), 2,2-dimethylcyclopropane (3) and tetramethylcyclopropane (4), are described. The spectral properties of these molecules demonstrate unambiguously the ring closed structure for cyclopropanones. The reactions of cyclopropanones with methanol, diazomethane and furan are employed as chemical means of characterization.

In this paper we describe an extensive structure–activity relationship study of this class of MDM2 inhibitors, which led to the discovery of 60 (AA-115/APG-115). t-Butylcyanoketen with bicyclo[2,2,1]heptadiene gives a tetracyclic adduct (2) by a formal [2 + 2 + 2] cycloaddition involving the keten carbonyl group; with isobutene the keten gives an “ene”-product (7).
