  Synthesis of novel pyrrolyl-indomethacin derivatives

  摘要：

  In the present work, we report the synthesis of the novel esters of indomethacin (IDMC) and an ester of reduced IDMC. For this purpose, 1DMC is covalently bound by using a spacer chain to the pyrrole (Py) in the 3-position. The innovative pyrrole-indomethacin (3-Py-IDMC) derivates show no cytotoxic effects in primary calvarial osteoblasts. The designed IDMC derivates have been studied because they could be injected locally as a component of polymeric micro-particles. In fact, the new 3-Py-IDMC derivatives will assure their further polymerization since the 2- and 5-monomer positions are free. (C) 2012 Elsevier Masson SAS. All rights reserved.

  The synthesis and characterization of various poly(pyrrole)s derived from (1H-pyrrol-3-yl)acetic acid are described. The stability and the conductivity of polymers from ester derivatives of (1H-pyrrol-3-yl)acetic acid with linear or branched alkyl chains, ether or thioether functions and polyfluorinated chains are evaluated. The electroactivity of these new conducting materials has been determined in both aqueous and organic media by cyclic voltammetry. Some of these polymers show ionoselective or enantioselective properties.

  Pyrroles having 3-substituent bearing the vinyl group with an ester or amide spacer were synthesized starting with 1-(phenylsulfonyl)pyrrole. Homopolymerization of the vinyl monomers thus synthesized provided cross-linked insoluble polymers, but their copolymers with 1-vinyl-2-pyrrolidone were soluble in common solvents.

  5,10,15,20-Tetraphenylporphyrins (Por) with a styrenic double bond-bearing substituent attached at the pyrrole residue with an amide or ester spacer were synthesized starting with 3-substituted pyrroles. Free radical copolymerization of ZnPor, prepared from Por, with 1-vinyl-2-pyrrolidone gave more favorable results than did Por.
