Results of studies of the nucleophilicaroylation of PhCO2Me, Me3CCO2Me and Ph2CO by several lowtemperature ArLi/CO systems (Ar =C6H5, p-CH3C6H4, -CH3C6H4, 2,6-(CH3)2C6H3 and 2,4,6-(CH3)3C6H2) are reported. The experiments with C6H5Li were unsuccessful and the best results were obtained with 2,6-(CH3)2-C6H3Li.

The 15.1 Mc/s 13C n.m.r. spectra of 21 substituted alkyl phenyl ketones, ArCOR, have been determined and the 13C chemical shifts measured. This group of compounds included examples of each of the ethyl, isopropyl, and t-butyl phenyl ketone series. derivatives with substituents in 2- and 6-positions has been synthesized. A number of compounds were investigated by dynamic NMR spectroscopy. Restrictedrotation was observed in 3-methyl-2-neopentylbenzoic acid and its derivatives. Thermodynamic and Arrhenius parameters were obtained for the restrictedrotation of the neopentyl group. The present invention relates to compounds which bind to Beta Trans-ducin repeat-containing protein (βTrCP), and modulate the activity of βTrCP. In particular, the invention relates to compounds which demonstrate optimised binding to PTrCP. The invention also relates to pharmaceutical compositions comprising such compounds and the use of such compounds as medicaments, specifically for the treatment of disorders associated with aberrant protein degradation, such as cancer. The preferred binding inhibitors are peptides derived from the motive DSGXXS, e.g. DEGFWE, DDGFWD and Succinyl-EGFWE.
