5-乙炔基-2'-脱氧胞苷是一种有机中间体,可通过将5-碘-2'3'5'-氧-三乙酰基尿苷(15 g,30.23 mmol,等当量)溶解于200 ml CH₂Cl₂∶(C₂H₅)₃N 1∶1 混合溶液中,并在氮气保护下加入(PPh₃)₂PdCl₂(1.59 g,2.27 mmol,等当量的七分之一),CuI(0.86 g,4.52 mmol,等当量的十分之二),以及三甲基硅基乙炔(6.24 g,8.8 ml,63.53 mmol,等当量的二倍)来制备。在氮气保护下常温搅拌20小时后,旋蒸除去溶剂,残余物用300 ml CH₂Cl₂溶解,并依次用水溶液(2×200 ml 10% EDTA二钠水溶液和200 ml 饱和食盐水)洗涤。水层再用200 ml CH₂Cl₂返萃取,合并有机层并旋干溶剂后进行柱层析分离,使用洗脱剂比例为PE∶EA 3∶1~1∶1,最终得到淡黄色泡沫状产物5-[(三甲基硅基)乙炔基]-2'3'5'-氧-三乙酰基尿苷11.4 g,产率为81%。
