Here we summarise our results for SmI2-induced 5-exo-trig to 8-exo-trig reductive cyclisations of suitably substituted indole and pyrrole derivatives. All precursors were easily prepared by simple N-alkylation or N-acylation of indole and pyrrole derivatives with the corresponding iodo alkanones, acid chlorides or lactones. The present disclosure provides pharmaceutical compositions, and their methods of use, where the pharmaceutical compositions comprise a prodrug that provides enzymatically-controlled release of a drug and an enzyme inhibitor that interacts with the enzyme(s) that mediates the enzymatically-controlled release of the drug from the prodrug so as to attenuate enzymatic cleavage of the prodrug. The disclosure provides pharmaceutical compositions which comprise an enzyme inhibitor and a prodrug that contains an enzyme-cleavable moiety that, when cleaved, facilitates release of the drug.
Synthesis of 4-Quinolones via Cyclocondensation of Substituted ortho-Amidoacetophenones: A Refit to the Camps Cyclization by Applying Trimethylsilyl Trifluoromethanesulfonate/Triethylamine
A modification of the classical Camps cyclization is described. A series of substituted 4-quinolone derivatives is prepared via trimethylsilyl trifluoromethanesulfonate/triethylamine induced cyclocondensation of substituted ortho-amidoacetophenones. The process shows a broad substrate scope and allows selective preparation of 2-aryl- and 2-alkyl-substituted 4-quinolones. Enantiopure starting materials react without loss of optical purity using the modified conditions. Subsequent transformations of the products involving preparation of a 4-quinolyl nonaflate and O-selective methylation are also described.
Functional group-selective poisoning of molecular catalysts: a ruthenium cluster-catalysed highly amide-selective silane reduction that does not affect ketones or esters
The addition of amines eliminates the catalytic activity of a trirutheniumcluster in the hydrosilane reduction of ketones and esters without affecting the rate of reduction of amides; selective reduction of the amide group in amido ketones and amido esters is accomplished.
