One-pot synthesis of functionalized pyrazolo[3,4-c]pyrazoles by reaction of 2-cyano-N-methyl-acrylamide, aryl aldehyde, and hydrazine hydrate

A simple and efficient procedure for the synthesis of novel 4-aryl pyrazolo[3,4-c]pyrazol-3(2H)-ones via a one-pot, three-component reaction between 2-cyano-N-methylacetamide, aryl aldehydes, and hydrazine hydrate in the presence of Et3N in DMF is reported. Products are obtained in good yields and their structures are supported by their spectroscopic data and combustion analysis.[GRAPHICS].

Reformatsky reaction of methyl 1-bromocyclopentane-1-carboxylic acid with 3-aryl-2-cyanoprop-2-enamides and their N-methyl derivatives afforded 10-aryl-6,8-dioxo-7-azaspiro[4.5]decane-9-carbonitriles and 10-aryl-7-methyl-6,8-dioxo-7-azaspiro[4.5]decane-9-carbonitriles, respectively.
