Structures of the ribose-methylatednucleosides 5,2′-O-dimethylcytidine (1), N4-acetyl-2′-O-methylcytidine (2), 2-thio-2′-O-methyluridine (3), and N2,N2,2′-O-trimethylguanosine (4) from the transferRNA of Sulfolobus solfataricus, Thermoproteus neutrophilus, and Pyrodictium occultum have been established, and verified by chemical synthesis.

Phosphorylated nucleotides for addition to nucleotide chains in the stepwise synthesis of oligonucleotides, e.g. on a polymeric support, are prepared in dioxane solvent and then lyophilized, to provide a stable, dry product ready for re-dissolving in reaction solvent. The lyophilized products are stable indefinitely and do not suffer from decreased reactivity.
