Compounds and compositions are presented that inhibit K-Ras, and especially mutant K-Ras. Certain compounds preferentially or even selectively inhibit specific forms of mutant K-Ras, and particularly the G12D mutant form.

Die vorliegende Erfindung betrifft ein dreistufiges Verfahren zum Herstellen von 4,4-Di-fluor-acet-essig-säure-ethyl-ester, worin im ersten Schritt 4-Chlor-4,4-di-fluor-acet--essig-säure-ethyl-ester mit Trialkylphosphiten der Formel (III) P(OR1)3 in welcher R1 für C1-C4-Alkyl steht, wobei die Reste R1 jeweils gleich oder verschieden sein können, zu Alkylphosphonsäureestern der Formel (IV) umgesetzt wird, welche im zweiten Schritt mit einem Amin der Formel (V) in welcher R2 und R3 unabhängig voneinander für Wasserstoff oder C1-C4-Alkyl oder ge-mein-sam für -CH2-CH2-O-CH2-CH2- stehen, zu Enaminen der Formel (VI) in welcher R2 und R3 die oben angegebenen Bedeutungen haben, umgesetzt werden, welche im dritten Schritt in Gegenwart einer Säure hydrolysiert werden.

The disclosure provides a process for the preparation of alkyl 3-difluoromethyl-1-methyl-1H-pyrazole-4-carboxylate and its analogs. The process includes a reaction workup method for Claisen condensation, wherein the enolate salt is acidified after removing remaining starting material and byproducts such as, ethanol and excessive ethyl acetate. The process also includes a method for completely drying alkyl difluoroacetoacetate and its analogs before use in the next step by reacting trialkyl orthoformate with the residual water. to promote the ring-closure reaction to produce the alkyl 3-difluoromethyl-1-methyl-1H-pyrazole-4-carboxylate. The process also includes effectively removing the regioisomer, alkyl 3-difluoromethyl-2-methyl-1H-pyrazole-4-carboxylate formed as a byproduct of the ring closure by a precipitation in a mixed solvent system and thereby eliminating the need for recrystallization of the final product.
