An efficient synthesis of a novel class of potent macrocylic renininhibitors exemplified by compounds 1 and 2, which involves the marcocyclization of 8 and 9 as the key step, is described. The macrocyclic design of renininhibitors 1 and 2 disclosed here incorporates (2R,3S)-3-amino-4-cyclohexyl-2-hydroxybutanoic acid (norACHPA) as the transition-state isostere. A wide variety of epoxides and aziridines were converted to the corresponding β-azido alcohols and β-azido amines with sodiumazide using Oxone® in aqueous acetonitrile. The reactions were highly regioselective and efficient with excellent yields at room temperature under mild reaction conditions.
Catalyst-Free Highly Regio- and Stereoselective Ring Opening of Epoxides and Aziridines with Sodium Azide Using Poly(ethylene glycol) as an Efficient Reaction Medium
Ring opening of epoxides and aziridines has readily been carried out at room temperature using poly(ethylene glycol) (PEG-400) as the efficient reaction medium to form the corresponding 2-azido alcohols and 2-azido amines, respectively, in excellent yields (95-99%) within 30-45 minutes and with high regio- and streoselectivity.
Fe3O4 nanoparticle-bonded β-cyclodextrin as an efficient and magnetically retrievable catalyst for the preparation of β-azido alcohols and β-hydroxy thiocyanate
A novel Fe3O4 nanoparticle-bonded β-cyclodextrin (β-CD@MNP) has been synthesized and characterized by scanning electron microscopy, vibrating sample magnetometry, and thermogravimetric analysis. It is found that the nanoparticles can be used as an efficient and magnetically retrievable nanocatalyst for the preparation of β-azido alcohols and β-hydroxy thiocyanates under ultrasound irradiation.
