制备方法与用途

在5 L三口反应瓶中,加入201.7 g 1,1-环丙烷甲酸乙酯、351.3 g DPPA、167.6 g 三乙胺和2.4 L MeCO。室温搅拌反应3小时后,加热至90℃回流反应3小时。通过TLC监控反应完全程度,自然冷却至室温后,在50℃下旋蒸除去MeCO。随后使用(EA 400 mL + NaCO3 400 mL)×2进行萃取,分出有机相并减压浓缩去除t-BuOH和EA,最终得到Boc-1-氨基环丙基甲酸259.3 g,产率为88.7%。
