    合成旋光性抗真菌唑TAK-187,2-[((1R,2R)-2-(2,4-二氟苯基)-2-羟基-1-甲基-3-(1H-1,建立了2,4-三唑-1-基)丙基] -4- [4-(2,2,3,3-四氟丙氧基)苯基] -3(2H,4H)-1,2,4-三唑酮。关键合成中间体2-[[(1R)-2-(2,4-二氟苯基)-2-氧-1-甲基乙基] -4- [4-(2,2,3,3-四氟丙氧基)苯基]-从(S)-乳酸的酯(11a,b)以立体控制的方式制备3(2H,4H)-1,2,4-三唑酮(8)。将该光学活性的苯乙酮衍生物8转化为一个碳延长的(1R,2S)-二醇7,然后使其与1H-1,2,4-三唑反应生成TAK-187。该新开发的路线适用于合成含咪唑酮或咪唑烷酮核的类似物(25a,b--28a,b)。

    New optically active antifungal azoles, 1-[(1R, 2R)-2-(2, 4-difluoro- and 2-fluorophenyl)-2-hydroxy-1-methyl-3-(1H-1, 2, 4-triazol-1-yl)propyl]-3-(4-substituted phenyl)-2(1H, 3H)-imidazolones (1, 2) and 2-imidazolidinones (3, 4), were prepared in a stereocontrolled manner from (1S)-1-[(2R)-2-(2, 4-difluoro- and 2-fluorophenyl)-2-oxi-ranyl]ethanols (15, 16). Compounds 1-4 showed potent antifungal activity against Candida albicans in vitro and in vivo, as well as a broad antifungal spectrum for various fungi in vitro. Furthermore, the imidazolidinones, 3b-e and 4d, e, were found to exert extremely strong growth-inhibitory activity against Aspergillus fumigatus.

    A cycloolefinic resin composition of the invention comprises a thermoplastic resin comprising an alicyclic olefin polymer and a specified stabilizer selected from among phenolic stabilizers, organic thioether stabilizers, organic phosphite stabilizers, ultraviolet absorbers and hindered amine stabilizers. Since this composition contains a specified stabilizer, it is excellent in thermal aging resistance and weatherability and can be preferably used as the resin composition for making an optical member. The thermoplastic resin may also be one comprising both of an alicyclic olefin polymer and a flexible polymer, whereby especially the mechanical strengths are improved and a resin composition containing said resin is preferably used in the manufacture of machine parts, electronics components and automotive parts.

    The cycloolefin resin compositions of the present invention comprise a thermoplastic resin containing an olefin polymer having an alicyclic structure, specific stabilizers selected from phenolic stabilizers, organic thioether stabilizers, organic phosphite stabilizers, UV-ray absorbers and hindered amine stabilizers. The cycloolefin resin compositions show excellent thermal aging resistance and weathering resistance by the use of specific stabilizers, and therefore they are favorably used for forming optical instrument parts. Furthermore, the thermoplastic resin may be prepared from an olefin polymer having an alicyclic structure and a flexible polymer or flexible polymers. The incorporation of the flexible polymer especially improves the mechanical strength of the resultant products, and therefore the resin compositions are favorably used for machine parts, electronic parts, automobile parts, etc.
