N-Fmoc-S-三苯甲基-D-半胱氨酸在常温常压下为白色固体状。
用途N-Fmoc-S-三苯甲基-D-半胱氨酸主要用于生物大分子多肽的合成中,例如可用于艾普替巴肽(eptifibatide peptide)的制备。此外,它也是生物大分子奥曲肽(octreotide)的关键合成中间体。
合成方法往真空干燥的反应瓶中加入9-(芴甲氧羰基氧)琥珀酰亚胺(Fmoc-OSu,1.75克,5.2毫摩尔),并用乙腈(11毫升)溶解。将氨基酸的前体Cys (Trt)-OH(1.89克,5.20毫摩尔)加入到碳酸钠水溶液中(600毫克,5.67毫摩尔,11毫升),再转移到Fmoc-OSu的溶液中。搅拌反应1小时后,在0度下用1M HCl将溶液酸化至pH 2,用二氯甲烷萃取该溶液三次,分离合并有机层,再用饱和的氯化钠水溶液洗涤合并的有机层,有机层用无水硫酸镁干燥,过滤混合物除去硫酸镁固体。得到的滤液在真空中浓缩即可得到产品N-Fmoc-S-三苯甲基-D-半胱氨酸。
