    An improved and scalable synthetic method of N-(5-(4-cyanophenyl)-3-hydroxypicolinoyl)glycine, a promising inhibitor of PHD2, is described. The optimized route staring from commercially available 5-bromo-3-nitropicolinonitrile takes only five chemical steps and has a dramatically increased total yield of 37.6% without chromatographic purifications.     The present invention relates to the field of pharmaceutical chemistry. In particular, the present invention relates to a class of alkynyl pyridine prolyl hydroxylase inhibitors (I). The experiments show that such a compound has good activity of inhibiting prolyl hydroxylase, and can enhance the generation and secretion of erythropoietin in cell or animal models, and thus can promote the generation of red cells, and can be used for the treatment or prevention of anemia, such as chronic kidney disease anemia, and ischemic diseases, including ischemic strokes, myocardial ischemia and other related diseases. The present invention also discloses a method for preparing such a compound.

    This invention provides methods for the treatment of anemia in patients with chronic kidney disease (CKD) using vadadustat (Compound 1), including methods suitable for conversion, correction, and maintenance therapy for patients. For example, methods described herein are durable, with efficacy observed for 24-52 weeks. Methods described herein can be particularly beneficial for patients converting from a previous anemia treatment comprising administration of an erythropoietin stimulating agent (ESA) such as darbepoetin alfa (DA), CKD patients on dialysis (e.g., peritoneal dialysis or hemodialysis), or CKD patients having certain hemoglobin (Hb) levels.
