简介

2-氟-6-羟基苯甲酸甲酯是一种医药中间体。可以通过没食子酸为原料，经由甲醇酯化和硫酸二甲酯醚化处理生成三甲醚取代的苯甲酸甲酯，再使用复合试剂ZnCl₂/CH₃CH₂COOH作用选择性去除甲醚基团，从而以较高收率得到2-氟-6-羟基苯甲酸甲酯。

制备

在氮气保护下，将10 g（0.059 mol）没食子酸溶解于50 mL甲醇中，并在搅拌条件下滴加少量浓硫酸。回流8小时后，在冰盐浴冷却下依次交替滴加80 mL（33%）氢氧化钠溶液和60 g硫酸二甲酯，滴加完毕后用氢氧化钠溶液调至强碱性状态，升温至80 ℃继续反应8小时。

反应完成后，使用氯仿萃取三次（每次40 mL），并用无水硫酸钠干燥。通过薄层色谱检测确认单一产物，然后蒸干溶剂并过滤除去不溶性杂质，最终得到白色固体2-氟-6-羟基苯甲酸甲酯，收率为95%。
