(S)-1-BOC-吡咯烷-3-甲酸又叫(S)-N-Boc-β-脯氨酸,是一种氨基酸衍生物。它可以由(S)-β-脯氨酸与Boc2O一步反应制备得到。有文献报道其可用于制备USP30抑制剂。
制备将152mg (1.32mmol) 的(S)-β-脯氨酸和350mg (1.58mmol) Boc2O 悬浮在1mL去离子水和3mL丙酮中。将混合物冷却至0℃,加入72mg (0.660mmol) 固体Na2CO3。反应3小时后,真空除去丙酮,并滴加20%(w/v)柠檬酸水溶液,直至pH值达到4。接着,将混合物用15mL水和15mL乙酸乙酯稀释。用水相萃取两次,每次使用10mL乙酸乙酯。合并的有机相经过Na2SO4干燥后浓缩。产物沉淀为白色固体,最终得到284mg (99%) 的(S)-1-BOC-吡咯烷-3-甲酸。
应用(S)-1-BOC-吡咯烷-3-甲酸可用于制备含有泛素C-末端水解酶30或泛素特异性肽酶30(USP30)抑制剂的化合物。
