1-苄基-4-哌啶甲酸乙酯及其衍生物是重要的药物中间体,具有很高的生物活性。该化合物可用来合成消炎药物类胰蛋白酶抑制剂,在受体水平上拮抗炎症介质的作用;此外,它还可用于合成抗肿瘤药、基质金属蛋白酶抑制剂和法尼基转移酶抑制剂。总体而言,1-苄基-4-哌啶甲酸乙酯及其衍生物不仅可用于合成GABA摄入抑制剂、抗肿瘤药物,还能合成生长激素促分泌素、消炎镇痛药物、心血管药物、促智药物、抗流感病毒药物和骨疾病药物等。
合成方法将20.01克(127毫摩尔)的异戊二酸乙酯与21.7毫升(0.15摩尔)三乙胺溶解在2升二氯甲烷中,冷却至0℃。逐滴加入18.2毫升(0.15摩尔)苄基溴。将混合物加热至室温并搅拌过夜。然后将悬浮液倒入饱和碳酸氢钠水溶液(100毫升),水层用二氯甲烷(2×100毫升)洗涤,合并的有机层用水盐(100毫升)清洗,用无水硫酸钠干燥、过滤,并真空浓缩。
通过柱色谱法(乙酸乙酯/己烷,1/3,体积比)纯化产物,得到1-苄基-4-哌啶甲酸乙酯27.40克,收率为87%。
