    Staudinger-Phosphonite Reactions for the Chemoselective Transformation of Azido-Containing Peptides and Proteins
    摘要:
    Site-specific functionalization of proteins by bloorthogonal modification offers a convenient pathway to create, modify, and study biologically active biopolymers. In this paper the Staudinger reaction of aryl-phosphonites for the chemoselective functionalization of azido-peptides and proteins was probed. Different water-soluble phosphonites with ollgoethylene substituents were synthesized and reacted with unprotected azido-containing peptides in aqueous systems at room temperature in high conversions. Finally, the Staudinger-phosphonite reaction was successfully applied to the site-specific modification of the protein calmodulin.

    Here we report and establish a facile synthetic method for these unprecedented sulfur-fused polycyclic aromatic hydrocarbons (S-PAHs) with symmetrical structures (C2-rectangle and D6h-hexagonal shape).     A series of fluorescent sensor molecules based on a phosphane sulfide derivative that is soluble in an organoaqueous solvent were designed and synthesized. The structure of the fluorophore has been optimized in order to have the best compromise in terms of solubility and photophysical properties. The obtained properties are in full agreement with quantum chemical calculations. A fluorescent molecular sensor containing one polyoxoethylene group has been synthesized and an efficient quenching upon mercury complexation has been observed. Finally, this sensing molecule has been introduced in a microfluidic chip in which fluorescence detection has been integrated. An efficient fluorescence response was observed upon mercury addition.
