    Novel heterocyclic thyromimetics are presented carrying carboxy-substituted benzofurans or sulfur containing heterocycles, as replacements for the amino acid side chain of T3. Potent agonists were identified in both series. SAR trends are examined and found to be mostly consistent with previously published thyromimetics. The lack of isoform selectivity demonstrated with isoform-selective transient THR transfection assays has been confirmed by corresponding in vivo studies. (C) 2007 Elsevier Ltd. All rights reserved.

    Die Erfindung behandelt substituierte Benzofuranderivate, Verfahren zu ihrer Herstellung sowie deren Verwendung als Azneimittel zur Behandlung von Alzheimer, Herz-/Kreislauf Erkrankungen bzw. Diabetes. Die Wirksamkeit der Verbindungen ist assoziiert mit einer Serumcholesterin absenkenden Wirkung. Formel (I), im welcher Z für O, S, SO, SO2, CH2, CHF, CF2 oder für NR8 steht, worin R8 Wasserstoff oder (C¿1?-C4)-Alkyl bedeutet, R?5¿ zusammen mit den beiden Kohlenstoffatomen des Phenylrings einen gegebenenfalls einfach oder zweifach, gleich oder verschieden substituierten, gesättigten oder ungesättigten Furanring bildet, R7 für Wasserstoff oder für Kation oder eine Gruppe steht, die unter physiologischen Bedingungen unter Bildung einer OH-Funktion abgespalten werden kann, wie vorzugsweise Alkyl oder Acetyl.
