  • Compositions comprising cleavable crosslinker and methods

    A composition is described comprising a polymer. The polymer comprises polymerized units derived from a cleavable crosslinking monomer. The cleavable crosslinking monomer comprises at least two free-radically polymerizable groups and at least one group having the formula —O—C(R2)(R3)—O—, wherein R2 and R3 are independently hydrogen, alkyl, or aryl. The composition has a Tg no greater than 50° C. In some embodiments, the composition has a storage modulus greater than 3×105 Pa at 25° C. and 1 Hz and may be characterized as a non-tacky polymer. In other embodiments, the composition is a pressure sensitive adhesive. Upon cleavage of at least a portion of the polymerized monomeric units derived from the cleavable crosslinking monomer, the composition exhibits a change in at least one physical property. In another embodiment, the composition comprises a polymer and fragments. The fragments comprise the reaction product of a free-radically polymerizable group bonded to a polymer chain and a pendent hydroxyl group. The composition has a Tg no greater than 50° C. Also described are articles, comprising the composition described herein (i.e. before and/or after cleavage); as well as methods of making the composition and methods of making articles.

    A pattern article comprising a copolymer having polymerized units derived from a cleavable crosslinking monomer, to which a pattern is imparted by the release of incipient acid from a thermal acid generator or photoacid generator. The selective release of the acid provides a pattern of high crosslink density and low crosslink density as result of the incipient acid cleaving a portion of the crosslinks.

  • Acid-cleavable ketal containing poly(β-amino ester) for enhanced siRNA delivery

    The safe and effective intracellular delivery of nucleic acids remains the most challenging obstacle to the broad application of gene therapy in clinic. Endosomal escape of nucleic acids is also a major barrier for efficient gene delivery. Ketal linkage is known to readily cleave at the acidic pH of endosomal compartments. Here, we report ketal containing poly(beta-amino ester) (KPAE) as an acid-cleavable non-viral siRNA delivery system. KPAE efficiently condensed siRNA into nanocomplexes with a diameter of similar to 150 nm, which are stable under neutral conditions but rapidly dissociate to release siRNA at acidic pH. KPAE had a buffering capacity due to the presence of secondary amines in its backbone, confirmed by acid-base titration. Moreover, the studies of confocal fluorescence imaging using calcein and Lyso-Tracker Red revealed that KPAE disrupted endosomes by colloid osmotic mechanism and "proton sponge" effects. Cell culture studies demonstrated that KPAE can deliver tumor necrosis factor-a (TNF-alpha) siRNA to lipopolysaccharide (LPS)-stimulated macrophages and significantly inhibit the expression of TNF-alpha. The results demonstrate that acid-cleavable KPAE has great potential as gene delivery systems based on its excellent biocompatibility, pH sensitivity and high gene delivery efficiency.   • Design of Asymmetric Particles Containing a Charged Interior and a Neutral Surface Charge: Comparative Study on <i>in Vivo</i> Circulation of Polyelectrolyte Microgels

    Lowering the modulus of hydrogel particles could enable them to bypass in vivo physical barriers that would otherwise filter particles with similar size but higher a modulus. Incorporation of electrolyte moieties into the polymer network of hydrogel particles to increase the swelling ratio is a straightforward and quite efficient way to decrease the modulus. In addition, charged groups in hydrogel particles can also help secure cargoes. However, the distribution of charged groups on the surface of a particle can accelerate the clearance of particles. Herein, we developed a method to synthesize highly swollen microgels of precise size with near-neutral surface charge while retaining interior charged groups. A strategy was employed to enable a particle to be highly cross-linked with very small mesh size, and subsequently PEGylated to quench the exterior amines only without affecting the internal amines. Acidic degradation of the cross-linker allows for swelling of the particles to microgels with a desired size and deformability. The microgels fabricated demonstrated extended circulation in vivo compared to their counterparts with a charged surface, and could potentially be utilized in in vivo applications including as oxygen carriers or nucleic acid scavengers.
