Numerous analogues of the naturally occurring antibiotic althiomycin have been synthesised exploiting both total- and semi-synthetic methodologies. The antibacterial activity of these derivatives has been determined in whole cell assays and indicates the natural product exhibits a restricted SAR. (C) 2002 Elsevier Science Ltd. Four analogs of an antibiotic althiomycin were synthesized in order to clarify the structure-activity relationships, especially with regard to two asymmetric centers and the aldoxime configuration. The antibiotic activity of de(hydroxymethyl) analog is comparable to that of the original antibiotic for Gram-positive bacteria.
