    Lipoxygenase Inhibitory Activity of Octyl Gallate
    摘要:
    Octyl gallate inhibited soybean lipoxygenase-1 (EC 1.13.11.12, type 1) with an IC50 of 1.3 muM. The inhibition of the enzyme by octyl gallate is a slow and reversible reaction without residual activity. The inhibition kinetics analyzed by Lineweaver-Burk plots indicates that octyl gallate is a competitive inhibitor, and the inhibition constant, K-I, was obtained as 0.54 muM. One molecule of octyl gallate scavenged six molecules of 1,1-diphenyl-2-picrylhydrazyl and inhibited autoxidative lipid peroxidation. In addition, octyl gallate was effective in preventing lipid peroxidation.

    A series of the gallate esters of n-alkanols (C1–C12) was examined to determine their inhibitory activities against hyaluronidase and collagenase. Hexyl, heptyl, octyl, nonyl, and decyl gallates inhibited both hyaluronidase and collagenase, and the most potent inhibitor was octyl gallate against both enzymes. Octyl 3,5-dihydroxybenzoate showed inhibitory effects on hyaluronidase, whereas collagenase was inhibited by octyl 3,4-dihydroxybenzoate.

The thermoplastic polymer composition according to the present invention is excellent in handling properties and melt-moldability, because it shows non-tackiness. By using the thermoplastic polymer composition according to the present invention, a molded article having high qualities can be produced with good productivity.

    Octyl gallate (3,4,5-trihydroxybenzoate) was found to possess antifungal activity against Saccharomyces cerevisiae and Zygosaccharomyces bailii, in addition to its potent antioxidant activity. Catechol moiety is essential to elicit this activity. The primary fungicidal activity of octyl gallate comes from its ability to act as a nonionic surface-active agent (surfactant). The length of the alkyl chain is not a major contributor but plays an important role in eliciting the activity. (C) 2001 Elsevier Science Ltd. All rights reserved.
