制备方法与用途

生物活性

Mavacamten (MYK-461, SAR439152) 是一种针对心肌肌球蛋白（cardiac myosin）的小分子调节剂，能够调节肥厚型心肌病中过度的肌节收缩。

体外研究

MYK-461通过抑制β-心肌肌球蛋白-S1的磷酸释放速率，降低稳态ATP酶的活性。这一作用不仅影响了肌球蛋白化学机械周期中的限速步骤（磷酸释放），还减少了myosin-S1头部的数量，在弱结合状态向强结合状态转变时，myosin-S1头部与肌动蛋白细丝的相互作用也随之减少。此外，MYK-461还能降低肌球蛋白与ADP-肌动蛋白结合的速度以及ADP从myosin-S1中释放的速度。

体内研究

在移植有人源杂合突变的肌球蛋白重链的小鼠模型中，长期给药MYK-461可有效抑制心室肥大、心肌细胞混乱和心肌纤维化的发展进程，并减少与肥厚和促纤维化相关的基因表达。
