Novel compounds and methods of using the compounds are provided herein. The compounds include novel carbon analogs of pyranose derivatives discovered to have Toll- like receptor 4 (Tlr4) inhibitory activity. The methods provide for treating infectious, inflammatory and post-traumatic disorders.

Methyl α-D-glucopyranoside, methyl α-D-mannopyranoside and, in particular, the corresponding α-D-glycopyranosyl chlorides readily undergo allylation with allylsilanescatalyzed by trimethylsilyltriflate or iodotrimethylsilane in a highly stereoselective mode.
Thiophenyl glycosides are converted to C-glycosides by reaction with allyl or methallyltri-n-butylstannane using both “one-electron” and “two-electron” procedures, which give different stereoselectivities in some cases.
