There is an unmetneed for the development of safe, effective and non-addictive analgesics for treating chronic and neuropathicpain. We synthesized novel quinoline-derived trifluoromethyl alcohols (QTAs), characterized and screened them for safety, analgesic activity and mechanism using zebrafish based behavioral and molecular studies. We have identified two potent analgesic QTAs that act by blocking

Abstract This paper demonstrated a new and economical methodology using Fe(ClO4)2·H2O as a catalyst for the direct C(sp3)–H functionalization of 2-methyl azaarenes via addition to trifluoromethyl ketones. The use of Fe salts as a Lewis acid catalyst has shown great potential as an accessible, affordable, and effective catalyst for the reaction. Under mild, optimized conditions, an extensive range of
Lewis acid-catalyzed Csp3–H functionalization of methyl azaarenes with α-trifluoromethyl carbonyl compounds

A Lewis acid promoted Csp(3)-H bond functionalization of methyl azaarenes with alpha-trifluoromethylated carbonyl compounds is described. Catalytic amounts of Yb(OTf)(3) provided a straightforward access to the corresponding trifluoromethylated alcohols in excellent yields up to 94% under mild reaction conditions.
