  为了提高甘草次酸 （GA） 的溶解度和降血糖活性，探索了新化合物的活性作用机制。通过将氨基葡萄糖 （GlcN） 添加到 GA 的 C-30 羧基中，合成了一种新型 2-（N-3-乙酰甘草乙酰基）–N-吡喃葡萄糖基-2-乙酰酰胺（化合物 9），并通过网络药理学和分子对接探讨了化合物 9 的降糖活性机制。结果表明，化合物 9 的溶解度优于 GA，化合物 9 的 α-葡萄糖苷酶抑制作用 （IC50 = 0.160 mmol/L） 优于 GA （IC50 = 0.381 mmol/L）。HepG2 胰岛素抵抗 （HepG2-IR） 模型发现，在 GA 和化合物 9 的帮助下，胰岛素抵抗细胞中的葡萄糖消耗会增加。网络药理学筛选了化合物 9 和疾病的 268 个靶点。蛋白质相互作用网络中的核心基因是表皮生长因子受体 （EGFR）、磷酸激酶 （SRC）、MAPK1、MMP2、mmp9 等，参与前列腺癌、

  Synthetic O-glycopeptides as model substrates for glycosyltransferases

  摘要：

  A new approach to O-glycopeptides of the glucosamine type is described. N-Urethane protected, peracetylated glucosamine is converted into its 1-thio (1-bromo) derivative and used for glycosylation of a variety of protected serine or threonine derivatives as acceptors. The urethane group is easily exchanged for the natural N-acetyl moiety and O-deacetylation is achieved with hydrazine/methanol. The resulting O-GlcNAc derivatives are subjected to an enzymatic galactosylation procedure using beta-1,4-galactosyluwsferase (EC 2.4.1.22) to furnish O-glycopeptides of the neolactosamine type.

  The conditions in catalysts and in reaction time for the reaction of protected α-D-glucosamine hydrochloride with different substituted benzaldehydes have been investigated in the presence of various inorganic and organic bases. Based on obtained results, the general procedure for the synthesis of Schiff’s bases from tetra-O-acetyl-β-D-glucosamine hydrochloride and substituted benzaldehydes has been optimized. Reaction yields were 28-80%. Azomethines 6a-i have the scavenging effect on the DPPH radical with values of IC50 of 50-75 μM.

  With respect to an enhanced solubility under physiological conditions, a carbohydrate-containing hypericin-based second-generation photosensitizer was prepared. Its photochemical properties were tested by means of the light-sensitized destruction of bilirubin IX alpha to be even better than those of the parent compound hypericin. Investigations on binding-interactions with DNA showed promising results as well.

  Prednisolone succinate glucosamine (PSG) conjugate, a prodrug for prednisolone, was synthesized and confirmed by NMR and MS spectrum. The stabilities of the prodrug in PBS (pH 2.50, 5.00, 7.20, and 7.89) were studied. Cytotoxicity and uptake assay of the prodrug were perfomed on HK-2 and MDCK cell lines. The results showed that compared with prednisolone, the PSG not only did not increase the cytotoxicity but also improved the uptake to 2.2 times of prednisolone by the cells. Thus, it indicated that glucosamine might be a potential carrier for kidney-targeting delivery of prednisolone. (C) 2011 Zhi Rong Zhang.
