Synthesis of New Aminophosphonothiophene Derivatives

The reaction of gamma,gamma'-diphosphonylketones 1 with active methylenenitriles and sulfur in basic conditions led to the new 5-amino-2(phosphonomethyl)-3-(phosphonoethyl)thiophenes 2. These compounds were then used in the Horner-Wadsworth-Emmons reaction to give 5-amino-2-alkenyl-3-(phosphonoethyl)thiophenes 3. The structure of all obtained products was confirmed by NMR (H-1, P-31 C-13) and IR spectroscopy.

Two synthetic methods leading to the new n , n '-diphosphonylketones 2 and 2 ' are reported. The first method involves the base-catalyzed addition of diethylphosphite to diarylideneketones. The second one utilizes the reaction of triethylphosphite and ethoxydiphenylphosphine with g , g '-bis(dimethylamino)ketone hydrochlorides.
