    A device adapted for containing a volume of growth medium and employed for detection of gas produced by a micro­organism or microorganisms putatively contained within a test sample and added to the growth medium, comprising a primary chamber, a dome defined by the top surface of the primary chamber and communicating therewith, the dome establishing a fluid level and adapted for collection of gas produced by a microorganism within the growth medium, and a fill section adja­cent to and communicating with one end of the primary chamber, the fill section spaced apart from the dome and extending above the fluid level, and further defining an opening for the addi­tion of a test sample is disclosed. The device may be sealed by means of a removable closure. An apparatus consisting of an array of substantially identical devices is also disclosed. In addition, methods of using the device and apparatus for detect­ing and estimating numbers of microorganisms, and for analyzing samples putatively containing a microorganism, are disclosed.

    This invention relates to a biological indicator derived from a composition comprising: a host organism comprising a spore forming bacteria; a reporter gene for producing an indicator enzyme; a regulatory gene; and a vehicle for inserting the reporter gene and the regulatory gene in the host organism; the host organism bearing a transposable genetic element in its genome for inserting an insertion sequence in the regulatory gene; the insertion sequence comprising a transposase, a pair of terminal inverted repeat sequences, and at least one open reading frame for expressing the transposase. The vehicle may be taken up by the host organism. The insertion sequence may be inserted in the regulatory gene. The host organism may undergo sporulation to form the biological indicator. A process and an apparatus for using the biological indicator are disclosed.

    A solid growth plating medium in which Shigella organisms will grow and form colonies in the medium, and substantially, other microorganisms are inhibited or their colonies are differentiated from Shigella organisms. In one embodiment of the invention, colonies produced by Shigella appear with the color of the plating medium, usually a clear off-white color, that can be readily observed. In another embodiment, the fact that Shigella boydii and Shigella sonnei produce the enzyme alpha-galactosidase, but most Shigella dysenteriae and Shigella flexneri strains do not, is utilized with a chromogenic substrate to produce colonies of these microorganisms of a distinguishing color.
