    藤椰茶作为预防和治疗许多慢性疾病的最有前景的膳食补充剂之一,受到广泛关注。游离脂肪酸在人体中作为必需营养素起着重要作用。首次使用对甲苯磺酸2-(萘-1-基-苯氨基)-乙基酯作为前柱荧光衍生化试剂,利用高效液相色谱-大气化学电离质谱(APCI/MS)技术,研究了藤椰茶中31种游离脂肪酸的含量。结果表明,低的检出限(0.52-1.53 ng mL-1)为游离脂肪酸的微量分析和评估提供了保障。实验的回收率在88.9%-103.6%之间。本研究的结果显示,藤椰茶中游离脂肪酸含量最高的月份是四月。共发现了13种游离脂肪酸,其中包括生命必需的C18:2w6、C18:3w3、w6和C20:4w6。所开发的方法展示了系统检测药用植物生物活性成分的强大潜力。

    A pre-column derivatization method using 2-(11H-benzo[a]carbazol-11-yl) ethyl 4-methylbenzenesulfonate (BCETS) as labeling reagent followed by HPLC with fluorescence detection and on line APCI/MS for the determination and quantitative analysis of fatty acids from safflower has been successfully developed. The fatty acids could be easily and quickly labeled by BCETS at 95 ºC in the presence of K2CO3 as catalyst in DMF within 35 min. The derivatives exhibited excellent fluorescence property with excitation and emission wavelengths of 272 and 505 nm, respectively. The method displayed good selectivity, sensitivity, reproducibility and applicability. Qinghai and Xinjiang safflower (QHS and XJS), which were planted in different places in Qinghai province, were analyzed by the established method. The result showed the contents of total fatty acids, saturated fatty acids and unsaturated fatty acids from Qinghai safflower sample were all much higher than that from Xinjiang safflower sample, especially for Qinghai safflower sample planted in Datong county. The main unsaturated fatty acids were C18:1, C18:2 and C18:3 and the dominated saturated fatty acids were C16, C18 and C20, respectively. This work would be helpful for the safety assessment and quality control of the two kinds of safflowers.

    The determination of trace plant growth regulator (PGR) has received more and more attentions in the field of phytophysiology and food safety. But the simple and sensitive method for simultaneously analysing multiple classes of PGR remains poorly investigated. In this study, a new pre-column fluorescence labelling method using 2-(11H-benzo[a]carbazol-11-yl)-ethyl-4-methylbenzenesulfonate (BCETS) as the labelling reagent has been developed for simultaneous determination of seven PGRs (i.e., indole-3-acetic acid, 3-indolybutyric acid, 3-indolepropionic acid, jasmonic acid, gibberellin A(3), 1-naphthylacetic acid and 2-naphthaleneacetic acid) by HPLC with fluorescent detection (FLD). The proposed method offered the LOD of 0.34-0.73 ng/mL for seven PGRs, which were significantly lower than the reported methods. The crude extract without complex pre-treatments and purification was directly labelled by BCETS and analysed by HPLC-FLD, which facilitates the high-throughput sample screening. This method was proven to be inexpensive, simple, selective, sensitive, accurate and reliable for trace PGR determination. (C) 2014 Elsevier Ltd. All rights reserved.

    Rattan tea regarded as one of the most promising dietary supplements for the prevention and treatment of many chronic diseases has gained extensive attention. Free fatty acids play an important role in human body as essential nutrients. Contents of 31 free fatty acids in Rattan Tea were firstly investigated using toluene-4-sulfonic acid 2-(naphthalene-1-yl-phenyl-amino)-ethyl ester as pre-column fluorescence derivatization reagent by RE-HPLC coupled with an atmospheric chemical ionization-mass spectrometry source (APCI/MS). The results indicated that lower values of LOD (0.52-1.53 ng mL-1) provided the guarantee for trace analysis and evaluation of free fatty acids. The experimental recoveries were in the range of 88.9-103.6 %. Results of the present study indicated the highest free fatty acids content of Rattan Tea was in April. Thirteen free fatty acids were found, including C18:2w6, C18:3w3, w6 and C20:4w6 essential to life. The developed method exhibited powerful potential for the systematic detection of bioactive components of medicinal plants.
